Runescape is coded in Java ( thats why you need to have java installed  ). Runescape uses a software render, so it can work in any browser and does not require powerful video cards.(Writing a software render is fun but you need to know your maths. However, its a good way to get maximum coverage for users)

In java you code for web using applets for example.
So if you want to do 3d in a browser, you have to look for a way to make your applets render 3d views.
There is Java3D, though I don't know much about it and if it works in applets/browsers.

There are also java openGL systems (Java binding for openGL LWJGL,
JOGL, etc ) which allow you to use openGL in your java programs.

Check this link out

So you have options. :D

Hope that helps

You can even use ShockWave and Flash I believe is trying to get more into 3D now and then, but I don't know much about the multilayer flexibility if you have knowledge of SQL syntax and a database.

You could also try Silverlight. I think it is in Beta 2 right now but it does let you code in C#.
